SS-31 (Elamipretide) is a water-soluble tetrapeptide with an alternating aromatic-cationic motif that concentrates in the inner mitochondrial membrane independently of membrane potential. Rather than acting on a receptor, it associates selectively with cardiolipin — the signature phospholipid of the inner membrane — which is what gives the compound its mitochondrial specificity.
Cardiolipin organises the respiratory supercomplexes and maintains cristae curvature, and its peroxidation is an early event in mitochondrial dysfunction. Research applications therefore include studies of electron transport chain coupling efficiency, cristae morphology under stress, reactive oxygen species generation at complexes I and III, and models of age-associated decline in mitochondrial bioenergetics.
