5-Amino-1-methylquinolinium (5-Amino-1MQ) is a cell-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), the cytosolic enzyme that catalyses transfer of a methyl group from S-adenosylmethionine to nicotinamide. By occupying the nicotinamide binding site, it interrupts the conversion of nicotinamide to 1-methylnicotinamide.
Because that reaction diverts nicotinamide away from the NAD+ salvage pathway and consumes cellular methyl donors, NNMT inhibition is studied as a lever on both NAD+ availability and methyl-group economy. Research applications include investigation of adipocyte metabolic regulation, sirtuin activity as a downstream consequence of altered NAD+ pools, polyamine flux, and the role of NNMT expression in metabolic tissue models.
